I can't really see it being Wonder Woman because (1) It's too obvious. (2) DC already has a bevy of lesbians. I can't really see having another one would really matter all that much, even if it would be the most iconic super heroine.
Martian Manhunter would seem to be a bad idea because (1) as an alien, and a barely humanoid one at that, he really shouldn't find humans attractive as all. Much like Phillip Jose Farmer said about Superman, he might as well be shagging apes. (2) His name is "Man Hunter". The jokes would just never end.
What I really want to know is why couldn't they have just revealed it in a story, and then see if the news organizations would report on it? Having press releases announcing this bold move to come just makes the whole thing feel crass.

Having it be an Earth 2 character's a bit of a cop-out, but it would be a way to have a Green Lantern or Flash making headlines without affecting upcoming media tie-ins.
That all sounds so cynical when I type it out like that...

That all sounds so cynical when I type it out like that... •• Cynical, hmmmmm-maybe. But a few years -- no, better make that DECADES! -- back, DC got the national press all hot and bothered with THIS… …neglecting to mention (to any but those with the sharpest eyes) that it was a FLASHBACK to the marriage of Superman and Lois of Earth 2.

If it really IS the Earth-2 Superman, it would be a way for DC to have a gay Superman, but still not the "real" Superman. Though it could be interesting to see how the writers portray a mature gay man. Earth-2 Superman is clearly older -- there's gray in his hair, after all -- so it wouldn't be like he'd be prancing about the bars or bed-hopping, one would hope. A stable relationship? Perhaps they'll landscape or garden together? Go to fabulous cocktail parties for non-profit organizations.... ...no, no! Maybe the storyline will be Superman's lover in a horrible health crisis and Superman won't be able to visit him in the hospital because they don't have marriage equality in that state...

Without getting into spoiler territory, I'll say it's very, very unlikely that it would be the Earth-2 version of any of the big three.
I'll also speculate that it's not going to be the Earth-2 version of anybody, since DC probably doesn't want to have a whole lot of qualifiers attached to whatever big press release they've got in mind, since Marvel already did that by killing off Ultimate Spider-Man, which seemed to result in confusion and indifference from the press.
I'd put money on a Flash or a Green Lantern if I were a betting man.
