The Gladiator was always more of a pathetic case to me than a true villain. So I'll go with the Owl.
However, Daredevil never really hit its stride until Frank Miller came to the book and introduced some truly compelling villains.
In hindsight, moving the Kingpin over from Spider-Man was one of the best creative moves Marvel ever made. Spider-Man had an abundance of great villains and didn't really miss the Kingpin, who was never going to surpass the Goblins, Doc Ock, etc. But in Daredevil, he became the evil mastermind/arch-villain that the book had previously lacked.
Miller also made Bullseye a central villain and that worked out great, too.
